Researcher sketching construct design in a notebook

Workflow

Design Wizard

Four questions, one construct. The wizard takes an organism, a heat target, a strategy preference, and an export format β€” and builds the complete specification in between. No expertise required at the front door.

Four steps

The wizard is the fastest path from β€œI want to engineer a spicy X” to a download-ready construct. The four steps each do one thing and move on.

Step 1

Pick your host organism

Choose from 65 organisms across seven plant families. The wizard shows the feasibility tier, the number of heterologous genes required, and the estimated timeline before you commit.

Step 2

Set your heat target

Drag a slider to a SHU target or type a specific value. The wizard runs a quick FBA check and tells you whether the target is within the host's modeled ceiling β€” and what happens to the flux if it is not.

Step 3

Choose a strategy

Three modes: Conservative (fewest edits, highest confidence), Aggressive (maximum flux, more edits), or Custom (pick individual targets from the pathway). For first-time users, Conservative is the recommendation.

Step 4

Export

Choose a format (GenBank, FASTA, IDT, Twist, SBOL, or ST26) and download the full construct package β€” sequence, construct map, CRISPR guides, primers, and protocol PDF β€” in one ZIP.

02

What the wizard builds for you

Each step in the wizard calls the same engines that expert users run manually. The organism selection loads the host’s metabolic model. The FBA check runs the flux solver against the target. The strategy selection generates CRISPR guide candidates and ranks them. The export calls the codon optimizer and constructs the annotation map. The only difference from the manual workflow is that the wizard makes the decisions that require domain knowledge β€” you supply the biology goal.

Host model
Genome-scale metabolic model pre-loaded for selected organism
FBA check
Real-time flux feasibility β€” ceiling computed before strategy selection
Guide design
Top-3 guides per edit target, pre-scored and pre-filtered
Codon optimization
Organism-specific codon table, GC smoothing, restriction site removal
Construct map
Full annotated map, SnapGene-compatible
Export package
ZIP with sequence, map, guides, primers, and protocol PDF

For teams with and without bioinformatics support

The wizard was designed for two different users: a molecular biologist who wants to run a quick feasibility check without writing a pipeline, and a business-side researcher who needs to know whether a target organism is worth pursuing before commissioning wet-lab work. Both get the same output β€” a fully specified construct β€” but the wizard does not require either of them to understand how the engines work.

Users who do want to understand the details can drop out of the wizard at any step and switch to the individual engine views β€” the Designer, FBA, CRISPR, or Codon Optimize pages β€” where all parameters are exposed.
