Glossary

Definitions for the pathway, platform, and licensing terms used across Scoville Splice. Each entry links to the related concepts you need to read it fully.

Agrobacterium tumefaciens

A gram-negative soil bacterium that naturally transfers T-DNA from its Ti plasmid into plant nuclear genomes. The most widely used plant transformation vehicle. Scoville Splice constructs are delivered as binary T-DNA suitable for standard Agrobacterium floral dip or callus co-cultivation.

See also: T-DNA, Binary vector

BCAT

Branched-chain amino-acid aminotransferase. Supplies the branched-chain precursors (valine, leucine) that feed the fatty-acid arm of the capsaicinoid pathway.

See also: KAS, Capsaicinoid

Binary vector

A two-plasmid Agrobacterium system for plant transformation. The 'helper' Ti plasmid supplies the vir genes needed for T-DNA excision and transfer; the 'binary' plasmid carries the T-DNA between left and right border repeats and can be moved independently into different Agrobacterium strains.

Bottleneck (pathway)

A reaction whose flux is lower than optimal and limits the overall yield of the target metabolite. Identified by FBA as reactions with zero flux when others in the same pathway carry positive flux. The FBA simulator highlights bottlenecks in amber in the flux table.

See also: FBA (Flux Balance Analysis), PAL (Phenylalanine Ammonia-Lyase)

CAI (Codon Adaptation Index)

A value from 0 to 1 measuring how well a coding sequence matches the codon preferences of a given host. A CAI of 1.0 means every codon is the host's most-preferred synonym. The codon optimizer raises CAI from the average of a naïve Capsicum sequence (~0.78) to near 1.0 for each target organism.

See also: Codon optimization

Capsaicin

C18H27NO3. The principal pungent alkaloid in chili peppers. Binds the TRPV1 receptor on sensory neurons and triggers a sensation of burning heat.

See also: Capsaicinoid, Dihydrocapsaicin, Nordihydrocapsaicin

Capsaicinoid

The family of fatty-acid amides of vanillylamine produced in the placental tissue of Capsicum fruit. The five major members — capsaicin, dihydrocapsaicin, nordihydrocapsaicin, homocapsaicin, and homodihydrocapsaicin — determine both the SHU and the burn character of a pepper.

See also: Capsaicin, Dihydrocapsaicin, Placental tissue

Capsicum

The genus of pepper plants, native to the Americas. Five domesticated species: C. annuum (bell, jalapeño), C. chinense (habanero, ghost, Reaper), C. frutescens (Tabasco), C. baccatum, C. pubescens.

See also: Capsicum chinense, Cultivar
