Zero-Bitter Brassica: TGG1/TGG2 Myrosinase Knockout for IFI-B Construct Specifications

Priyanca Ford · ORCID 0000-0003-0395-1752 · Scoville Splice LLC · 2026

Abstract

Elimination of glucosinolate-derived bitterness in Brassica species via CRISPR-SpCas9 knockout of TGG1 and TGG2 (thioglucoside glucohydrolases / myrosinases), preventing hydrolysis of glucosinolates into bitter isothiocyanates, nitriles, and epithionitriles. Demonstrated across broccoli, Brussels sprouts, and kale. Construct specifications target TAS2R38 suppression for consumer and commercial bitterness reduction applications. PDF and full specifications in preparation.
