Order the Kit for Your Construct

Everything you need to take a Scoville Splice construct from a FASTA file to a living plant. We don’t sell physical kits — we give you the exact reagents list, vendor links, and protocol. You order directly from the suppliers at standard pricing.

Step 1 — Download your construct files

Each product page has a ZIP download containing: the codon-optimized FASTA for your target organism, a construct map (SnapGene-compatible), CRISPR guide RNA spacer sequences, primer recommendations, and a protocol PDF. This is everything a synthesis vendor or a competent cloning protocol needs.

Step 2 — Order synthesis

Submit the guide RNA spacer sequence (20 nucleotides, listed in your CRISPR guide file) to IDT for Alt-R gRNA synthesis. Typical cost: $15–20 per guide. For the full coding sequence, submit the codon-optimized FASTA to Twist or GenScript. Cost for a 3,000 bp Pun1 cassette: ~$270 at Twist pricing.

Step 3 — Choose a backbone

The construct FASTA is backbone-agnostic — it defines the sequence you want expressed, not the delivery vehicle. Choose a backbone based on your plant host, Cas protein preference, and cloning method (Golden Gate vs. Gibson). Three recommended options are listed in the Vendors tab.

Step 4 — Transform and validate

From here, follow the seven-step transformation protocol. The protocol PDF in your ZIP download has organism-specific notes for Capsicum, Solanum, and the five most common non-pepper hosts.
