Can you engineer rice to be spicy?
Yes, but it is a Tier 4 Frontier project โ one of the harder designs in the catalog. Rice sits far from Capsicum in evolution and does not supply any of the pathway on its own. The template is Golden Rice: build the pathway elsewhere, carry it over as a complete module.
What makes rice hard
Chili pepper is a dicot in the Solanaceae; rice (Oryza sativa) is a monocot in the Poaceae. The two lineages diverged more than 140 million years ago. That evolutionary distance shows up in three practical ways. First, rice has no detectable background flux through the phenylpropanoid-to-vanillin arm that supplies the amine half of capsaicin. Second, rice endosperm โ the edible part of the seed โ is dedicated to starch and storage protein; its branched-chain acyl-CoA pool is small. Third, codon optimization from Capsicum to Oryza is non-trivial because the two have different preferred codon usages and different GC content.
The Golden Rice template
Golden Rice, demonstrated by Potrykus and Beyer in 2000, is the cleanest precedent in cross-species crop engineering. The project reconstituted the ฮฒ-carotene biosynthesis pathway in rice endosperm by carrying over two genes โ psy from daffodil and crtI from the soil bacterium Pantoea ananatis โ under an endosperm-specific promoter. The result was a rice grain with provitamin A sufficient to address vitamin A deficiency in populations whose diet is dominated by white rice. The project took decades to clear regulatory review but is now approved in several countries.
The Golden Rice recipe generalizes: identify the smallest set of genes needed to reconstitute the target metabolite, codon-optimize them for the host, drive them from a tissue-specific promoter, and deliver as a single integrated T-DNA cassette.
What the construct looks like
Our Tier 4 rice design is a six-gene cassette: pAMT, Pun1, two upstream fatty-acid branch enzymes, a phenylpropanoid shunt that feeds vanillin production, and a selection marker. All six are codon-optimized for Oryza sativa and driven from an endosperm-specific globulin promoter. Delivery is through Agrobacterium transformation of calli, with regeneration into whole plants selected on the marker.

What SHU is realistic?
Our flux-balance ceiling for rice endosperm is approximately 20,000 SHU, limited by the small branched-chain acyl-CoA pool. In practice, Tier 4 rice cultivars target 500โ5,000 SHU โ a mild background heat that fits dishes where rice is the base rather than the condiment: biryani, jollof, arroz rojo, kimchi fried rice.
Timeline and licensing
Rice cultivars in the catalog ship as designs today. Taking a Tier 4 design from sequence to field-ready seed requires a multi-year wet-lab and regulatory process on the licensee's side, with Scoville Splice as design partner. The relevant license tier is Genome. See the rice organism hub for the current product line.
