How Do Taste Receptors Work?
Flavor perception is mediated by distinct protein families: G protein-coupled receptors (GPCRs), ion channels, and ionotropic receptors. Each binds a different class of molecule and generates a different neural signal. Engineering flavor means engineering ligands for these proteins.
Sweetness: TAS1R2/TAS1R3
Sweetness is detected by a heterodimer of two GPCRs: TAS1R2 and TAS1R3. The dimer has multiple binding sites — the Venus fly-trap domain in TAS1R2 binds sugars and artificial sweeteners, while the cysteine-rich domain binds brazzein and miraculin. When a ligand binds, the receptor activates gustducin (a G protein), triggering a cAMP/IP3 cascade that closes K⁺ channels and depolarizes the taste cell.
The IFI-S axis (sweetness) scores compounds relative to sucrose at receptor saturation, normalized to a 1–100 scale. Engineered constructs that upregulate the trehalose pathway or accumulate thaumatin are scored on this axis.
Umami: mGluR1 and T1R1/T1R3
Umami has two receptor pathways. The metabotropic glutamate receptor mGluR1 binds L-glutamate (and its synergistic enhancers, 5'-IMP and 5'-GMP), while the TAS1R1/TAS1R3 heterodimer serves as a broader "savory" receptor activated by L-amino acids. Both signal through separate downstream cascades, and their combined activation produces the characteristic mouthfilling, savory quality. The IFI-U axis quantifies this via mGluR1 binding affinity.
Bitterness: TAS2R Family
Humans express ~25 bitter taste receptors (TAS2Rs), each with a different ligand specificity. Cruciferous vegetables activate TAS2R38 through glucosinolate hydrolysis products (isothiocyanates). Coffee activates TAS2R43. Quinine activates multiple TAS2Rs simultaneously. Because these are all GPCRs signaling through the same downstream pathway, the aggregate bitter signal is the sum of activation across all expressed receptors.
The IFI-B axis targets TAS2R38 specifically — measuring bitterness attenuation in constructs that knock out glucosinolate pathway genes in brassicas.
Heat: TRPV1
Heat (pungency) is not a classic taste — it's a chemesthetic sensation mediated by TRPV1 (Transient Receptor Potential Vanilloid 1), a non-selective cation channel on sensory neurons. TRPV1 is activated by temperatures above ~43 °C, by acidic pH, and by chemical capsaicinoids. Capsaicin binding shifts the thermal activation threshold to room temperature, generating the same "burning" signal as physical heat.

Cooling: TRPM8
Cooling is mediated by TRPM8 (Transient Receptor Potential Melastatin 8), another TRP channel normally activated by temperatures below ~25 °C. Menthol, eucalyptol, and icilin activate TRPM8 at room temperature — the same mechanism that makes peppermint feel cool. The IFI-C axis quantifies TRPM8 activation, relevant for constructs engineering menthol content in herbs, peppermint, or basil.
Kokumi: CaSR
Kokumi (sometimes "mouthfulness" or "continuity") is an emerging category mediated primarily by the calcium-sensing receptor (CaSR), a GPCR expressed on Type I taste cells. Glutathione, γ-Glu-Val-Gly, and other γ-glutamyl peptides activate CaSR and amplify the perceived intensity, roundness, and persistence of sweet, salty, and umami signals without contributing flavor themselves. The IFI-K axis quantifies CaSR activation — particularly relevant for fermented ingredient constructs.
Astringency: Salivary Protein Precipitation
Astringency is not mediated by a single receptor. Instead, polyphenols (tannins, ellagitannins, catechins) cross-link proline-rich salivary proteins, causing them to aggregate and creating a rough, drying sensation. The IFI-A axis measures this via salivary protein precipitation assay — relevant for constructs targeting polyphenol profiles in grapes, pomegranates, or tea.
The Iyengar Flavor Index
The Iyengar Flavor Index (IFI) normalizes activation across all eight receptor systems — IFI-H, IFI-C, IFI-S, IFI-U, IFI-B, IFI-K, IFI-A, and IFI-P (sharp, TRPA1-mediated) — onto a common 0–100 scale using receptor saturation as the normalization reference. This allows a single construct to be described by an eight-dimensional flavor vector rather than a single number, enabling precise specification and comparison across any organism, tissue, or delivery platform.
